In a banana republic, way south of the Texas border, a dumb-Dora American girl, Norma (Olive Borden), lets her ruby-red lips promise more than she is willing to deliver, and she finds herself a prisoner in a notorious dance-hall/brothel. But her American aviator boyfriend, Barry Blake (John Boles), is flying to her rescue. He does just that but, alas, they are quickly captured by a gang of outlaws. Possibly the many expensive pieces of jewelry she has gathered from the many male friends she has made along the way, including El Presidente, captured the outlaws' attention.

Larry Nelson, paroled from prison after serving nearly half of his thirty-year sentence, is determined to not fall into the clutches of the law again, and takes a quiet job at a country sanitarium. Thete, he meets and falls for a nurse, Charlotte Maynard, and he knows the only way to enter her web is to have a lot of money, for Miss Maynard is somewhat of a gold-digger.

This one starts differently but, in the end, it is another version of Robert Emmett Tansey's oft-used plot of "employing bad guys as good guys to help the good-good guys capture the bad-bad guys." The warden of the Desert Wells Penitentiary asks Tex Reed and Slim to check the series of bank robberies which have been committed by escaped convicts. Lockwood, head of an opposing political machine, is behind the escapes and robberies, and the escapes are being planned by Red, a convict. Tex trails the next escapee but the hang shoots the man before Tex can question him. Jimmy, brother of Tex's girl friend Mary, is set up, by the gang, to be killed while robbing a bank by Carter who will collect a reward for shooting him. Jimmy is wounded but not killed and Tex arrests him to keep him safe. The gang now wants to get rid of Tex, so they send Red, dressed as a prison guard, with a fake message from the Warden for Tex.

Cheyenne Harry, owner of the biggest cattle ranch in his corner of the west, is having trouble with John Merritt, a land-grabbing Chicago meat-packer. He leaves the house chagrined but more determined than ever to get justice from Merritt.

Cullen has hired Tom to try and stop the robberies on his railroad. Knowing Cullen's secretary Holt is tipping off the gang, Tom works undercover by posing as a highwayman. To help him bring in the gang he enlists the help of the hobo DeLuxe Harry.

The son of Sheriff Clay Hartley, of the frontier town Elder, has gotten into bad company and hangs out with an outlaw gang in which, Collins, owner of the Golden Rule Saloon, is the secret head. Sheriff Hartley suspects him, but has been unable to gather the needed evidence. Collins instructs his gang, including young Hartley, to hold up the stagecoach on its return trip from Missionary Flats and take the cargo of gold dust it is carrying. Sheriff Hartley is notified of the planned holdup by one of his deputies who has been spying on Collins, and organizes a posse. A deputy-sheriff is killed in the ensuing gunfight between the lawmen and the outlaws, but Deputy Joe Larkin, pursues and captures Clay Hartley Jr. The latter is quickly tried and convicted of the killing of the deputy, and sentenced to be hung. Sheriff Hartley has only a few hours to prove his son was not the killer. He enlists the aid of Collins' step-daughter, Joan, who is in love with Hartley's son.

A dying Jack makes Bob and Flash promise not to tell his sister that he was an outlaw. When Bob confronts Flash with his muffler found at the stage holdup, Flash tells Mary that Bob killed her brother. Believing he can now marry Mary, he plans one more robbery. But the jealous Tiana overhears and runs for the Sheriff.

Charley Gray is about to be released from the state penitentiary after serving a long term for the robbery of a government gold shipment. The gold was never recovered, so the Texas Ranger chief has Ranger Panhandle Perkins planted in the prison as Charley's cell-mate in the hopes Charley will tell him where the loot is buried. Charley has a map of the location but is afraid it may be discovered so, while Panhandle is asleep, he draws a copy of it on the sole of Panhandle's foot. Charley then destroys the map but intends to keep "Panhandle" close to him upon their release from prison. Charley makes Panhandle accompany him back to the town where the rest of the hold-up gang is holed up. They go to the saloon owned by Steve Martin, also a member of the hold-up gang, but Charley was the one who buried the loot before he was captured and Charley has no intentions of divulging the location of the gold. With his father accused of murder, Tom heads after the real murderer who lives in a town of outlaws where no one is allowed in or out. To gain entry he poses as an escaping outlaw with his sidekick Banty posing as the pursuing lawman. This lets Tom join the gang but there is trouble later when Banty gets caught and sentenced to die.

Not knowing he has just been pardoned, Tim Benton (Tim McCoy, Texas Cyclone) escapes from prison with his cellmate, Red Larkin (Matthew Betz, The Wedding March), a dangerous killer. Disguised as the town's lawman, Tim sets off for Silver City to take back money that's rightfully his and hopefully clear his name. But Red has plans of his own and wants the money for himself. Cheyenne Harry tries to help his outlaw friend Padden evade arrest after Padden has drunkenly shot another man. In the end, the two mismatched friends fight it out, leaving Padden dead. In a romantic subplot, Harry's fiancée Alice leaves him, but finally returns.

Johnny Mack Brown's Universal western series was drawing to a close when Cheyenne Roundup was released in mid-1943. Brown is herein cast in a dual role, as honest Gils Brandon and his less-than-honest brother Buck. Pursued by lawman Steve Rawlins (Tex Ritter), Buck tries to pass himself off as the upright Gils.
